摘要：

目的探讨RECK和MMP-2基因在骨肉瘤中的表达及其临床意义。方法采用荧光实时定量PCR（RT-PCR）对我院2001年至2006年45例骨肉瘤手术切除标本中RECK mRNA、MMP-2 mRNA进行检测，同时结合患者的性别、年龄、转移、复发等临床病理特征，分析其表达的临床意义。结果 45例骨肉瘤组织中RECK mRNA和MMP-2 mRNA的相对表达量分别为38.15±14.15和65.00±22.86，两者表达呈负相关（r=-0.740，P=0.000）。RECK mRNA和MMP-2 mRNA表达与性别、年龄、肿瘤部位、组织亚型无关（P＞0.05），与肿瘤复发、转移及5年生存期显著相关（P＜0.05）。结论 RECK mRNA和MMP-2 mRNA表达与骨肉瘤转移、复发及预后相关，可作为预测骨肉瘤转移、复发及预后的分子指标。

Abstract:

Objective To explore the expressions of RECK mRNA and MMP-2 mRNA in osteosarcoma and their clinical significance. Methods RT-PCR was used to determine the expressions of RECK mRNA and MMP-2 mRNA in 45 osteosarcoma tissues from Nanjing General Hospital of Nanjing Military Command. Results The relative quantitative expressions of RECK mRNA and MMP-2 mRNA were 38.15±14.15 and 65.00±22.86 in osteosarcoma tissues. There was a negative relationship between two genes （r=-0.740, P=0.000）. The expressions of RECK mRNA and MMP-2 mRNA were correlated to metastasis, recurrence and prognosis（P＜0.05）, but they were not related to age, gender, tumor location and subtype（P＞0.05）. Conclusion The expressions of RECK mRNA and MMP-2 mRNA are correlated with recurrence，metastasis and prognosis. They can be relevant biomarkes in predicting metastasis, recurrence and prognosis of osteosarcoma patients.
