吗啡对人肺腺癌细胞A549侵袭与迁移的影响及机制研究

摘要: 目的 探讨吗啡对人肺腺癌细胞A549侵袭和迁移能力的影响。方法 人肺腺癌细胞A549培养板接种培养24 h后,分别加入生理盐水(对照组)、0.3 μg/ml吗啡、3 μg/ml吗啡和30 μg/ml吗啡孵育48 h,采用Transwell法检测细胞侵袭能力,细胞划痕实验检测细胞迁移能力,Western blotting检测基质金属蛋白酶-2(MMP-2)、MMP-9、Ezrin和Fascin蛋白的表达。结果 与对照组比较,3 μg/ml和30 μg/ml吗啡组处理A549细胞48 h后细胞侵袭数增加,差异有统计学意义(P<0.05);与对照组比较,3 μg/ml和30 μg/ml吗啡组处理A549细胞48 h后细胞迁移率增加,差异有统计学意义(P<0.05);与对照组比较,30 μg/ml吗啡作用A549细胞48 h后,MMP-2、MMP-9、Ezrin和Fascin蛋白表达上调,差异有统计学意义(P<0.05)。结论 吗啡可增强人肺腺癌细胞A549的侵袭和迁移能力,且呈剂量依赖性;其机制可能与MMP-2、MMP-9、Ezrin和Fascin表达上调有关。

Abstract: Objective To investigate the effects of morphine on the invasion and migration of human lung adenocarcinoma A549 cells line. Methods Human lung adenocarcinoma A549 cells were cultured in vitro for 24 hours and were randomly divided into 4 groups: control group, 0.3 μg/ml morphine group, 3 μg/ml morphine group and 30 μg/ml morphine group. The cells in the control group were treated with normal saline, whereas cells in the morphine groups were treated with morphine at different concentrations(0.3, 3, and 30 μg/ml). The invasion of cells was determined by transwell assay. The migration of cells was determined using the wound healing assay. The levels of matrix metalloproteinase-2(MMP-2), MMP-9, ezrin and fascin proteins were assessed by Western blotting analysis. Results Compared with the control group, morphine treatment increased tumor cell invasion and migration abilities at concentrations above 3 μg/ml(P<0.05). The expressions of MMP-2, MMP-9, ezrin and fascin proteins were significantly increased compared with the control group after treatment of morphine at the concentration of 30 μg/ml for 48 hours(P<0.05). Conclusion Morphine increase tumor cell invasion and migration of human lung adenocarcinoma A549 cells line in a dose-dependent manner, which may be associated with the upregulation expressions of MMP-2, MMP-9, ezrin and fascin proteins.
