肝癌组织中STOML2和TRIM25的表达及其临床意义

摘要: 目的 探讨肝细胞癌(HCC)组织中Stomatin样蛋白2(STOML2)和三基序蛋白25(TRIM25)的表达水平及其与临床病理特征的关系。方法 采用免疫组化EnVision二步法检测75例HCC组织和61例癌旁正常肝组织中STOML2和TRIM25的表达强度,根据细胞内染色强度和阳性细胞数分级,分析STOML2、TRIM25的表达与HCC临床病理特征的关系及两者的相关性;同时采用Kaplan-Meier法分析STOML2、TRIM25不同表达强度的总生存期(OS)。结果 STOML2和TRIM25在HCC组织中的高表达率分别为68.0%(51/75)和64.0%(48/75),均高于癌旁组织的24.6%(15/61)和37.7%(23/61),差异有统计学意义(P<0.01);STOML2表达与肿瘤大小、分化程度、BCLC分期、门静脉癌栓和肝外转移有关,而TRIM25蛋白表达与治疗前甲胎蛋白(AFP)、分化程度和腹水均有关(P<0.05),且STOML2与TRIM25表达呈正相关(r=0.379, P=0.001);TRIM25高表达者的中位OS为21.0个月,低于低表达者的36.5个月(P<0.05);但STOML2不同表达水平患者的中位OS的差异无统计学意义(P>0.05)。结论 STOML2和TRIM25在HCC组织中呈高表达,两者联合检测可用于评估肝癌的恶性表现、分化情况及肝转移程度,且TRIM25水平可能有助于生存预后的评估和预测。

Abstract: Objective To explore the expressions of stomatin like protein 2(STOML2) and tripartite motifcontaining 25(TRIM25) in the hepatocellular carcinoma(HCC) tissues as well as their relationship with clinicopathological characteristics. Methods The expressions of STOML2 and TRIM25 in 75 cases of HCC and 61 adjacent normal liver tissues were detected by immunohistochemical EnVision two-step method. The results were graded according to the intracellular staining intensity and the number of positive cells. Then the relationship of STOML2, TRIM25 expressions and the clinicopathological characteristics of HCC and the correlation of two proteins were analyzed. Meanwhile, the KaplanMeier survival curve was used to analyze the overall survival(OS) of HCC patients with different intensity of TRIM25 and STOML2. Results The over-expression rates of STOML2 and TRIM25 in HCC tissues were 68.0%(51/75) and 64.0%(48/75), both higher than 24.6%(15/61) and 37.7%(23/61) of the para-cancer tissues(P<0.01). The expression of STOML2 was related to tumor size, differentiation degree, BCLC staging, portal tumor thrombus and extrahepatic metastasis, while the expression of TRIM25 was associated with the AFP before treatment, differentiation degree and ascites(P<0.05). STOML2 was positively correlated with TRIM25(r=0.379,P=0.001). The median OS of patients with highlevel TRIM25 was 21.0 months, lower than 36.5 months of patients with low-level TRIM25(P<0.05), but there was no significant difference of median OS between patients with different levels of STOML2(P>0.05). Conclusion STOML2 and TRIM25 are highly expressed in HCC tissues and the combined detection of STOML2 and TRIM25 should probably be helpful for the evaluation the malignancy and differentiation of HCC. TRIM25 may be useful for the assessment and prediction of prognosis of HCC.
