Expression of SphK1 in gastric cancer and its clinical significance

Abstract:

ObjectiveTo investigate the role of Sphingosine kinase-1(SphK1) in the occurrence and progression of gastric cancer, and its correlation with vascular endothelial growth factor(VEGF). Methods Quantitative realtime reverse transcriptionPCR(qRTPCR) assay was used to determine the expression of SphK1mRNA in 31 cases human gastric cancer tissues and paracarcinoma tissues. Immunohistochemical method(SP) was used to detect the expression of SphK1 and VEGF protein. Results The expression of SphK1mRNA in gastric cancer and corresponding paracarcinoma tissues was 2.77±0.50 and 1.02±0.23(P<0.05). The high expression rate of SphK1 protein in the gastric cancer tissues was 71.0%(22/31), higher than 38.7%(12/31) in the corresponding paracarcinoma tissues(P<0.05). The expression of SphK1 in gastric cancer tissues was correlated with the depth of infiltration, TNM clinical stage, lymph node metastasis and vessel invasion, but not with age, gender and differentiation. The high expression rate of VEGF protein in the gastric cancer tissues was 64.5%(20/31). The expression of SphK1 protein were positively related with VEGF protein in gastric cancer(r=0.565, P<0.05). ConclusionSphK1 highly expresses in gastric cancer, and promote the occurrence and progression of gastric cancer in correlation with VEGF.
