摘要: Wnt信号通路在肿瘤细胞的增殖、分化及发生中发挥了重要作用,其异常激活是许多肿瘤的重要特征。作为分泌性Wnt信号通路的拮抗因子Dickkopf相关蛋白-3(DKK-3),深入研究其生物学功能以及抑制Wnt信号通路的机制,对肿瘤诊断、治疗及预后具有重要意义。本文就DKK-3的生物学作用,参与肿瘤不同阶段的发展,以及DKK-3表达/DKK-3启动子甲基化作为肿瘤生物标志物和分子靶点的潜在临床价值作一简要综述。

Abstract:

The Wnt signaling pathway plays an important role in cell proliferation, differentiation and carcinogenesis. Aberrant activation of the Wnt signaling pathway is a major trait of many human tumors. Dickkopf-related protein 3(DKK-3), an antagonist of Wnt signaling pathway, is involved in different stages of tumor development. The research on the biological function of DKK-3 and the mechanism of inhibition of the Wnt signaling pathway may have important significance of tumor diagnosis, treatment and prognosis. In this paper, we reviewed the biological role of DKK-3, the function of DKK-3 in different stages of tumor development, and the potential clinical value of DKK-3 expression and DKK-3 promoter methylation as a tumor biomarker and molecular target.
