氟他胺对乳腺癌HCC1937细胞的增殖抑制作用及机制研究

摘要:

目的 探讨氟他胺对乳腺癌HCC1937细胞增殖和凋亡的影响及可能的作用机制。方法 采用四甲基偶氮唑蓝(MTT)法检测不同浓度(5、10、20μmol/L)的氟他胺对人乳腺癌HCC1937细胞体外生长的抑制作用;Annexin V-FITC/PI流式细胞仪检测细胞凋亡情况;蛋白印迹法(Western blotting)检测核因子κB(NF-κB)和半胱天冬酶(Caspase-3)的蛋白表达水平。结果 不同浓度氟他胺作用于HCC1937细胞12、24、36、48h后,细胞生长受到抑制且呈时间和剂量依赖性。不同浓度氟他胺作用HCC1937细胞24、48h后,可显著促进细胞凋亡,并呈时间和剂量依赖性。氟他胺作用HCC1937细胞24h后,随着药物浓度的增加,NF-κB蛋白表达水平逐渐下降,而Caspase-3蛋白表达水平逐渐升高。结论 氟他胺能够抑制HCC1937细胞增殖和促进其凋亡,其作用机制可能是通过上调Caspase-3和下调NF-κB蛋白表达实现的。

Abstract:

Objective To investigate the potential antiproliferative and pro-apoptotic effects of flutamide on human breast cancer cell line HCC1937 in vitro and explore its possible anti-cancer mechanism. Methods Different concentration of flutamide(5,10,20μmol/L) were added to HCC1937 cells. The MTT assay was performed to reveal the inhibitory effect on cell proliferation. The cell apoptosis was determined by flow cytometry using annexin V-FITC/PI staining and the expressions of nuclear factor(NF-κB) and caspase-3 were examined by Western blotting analysis. Results Flutamide with different concentration treatment inhibited the proliferation and induced the apoptosis of human breast cancer cell line HCC1937 in a time- and dose-dependent manner. The expression of NF-κB protein decreased and caspase-3 protein increased after flutamide treatment for 24h. Conclusion Flutamide can inhibit the proliferation of human breast cancer cell HCC1937 and induce its apoptosis,with the possible mechanisms of dysregulation of NF-κB and caspase-3 expression.
