Cyclin D1、PTEN蛋白在甲状腺乳头状癌中的表达及临床意义

Expression of Cyclin D1 and PTEN protein in papillary thyroid carcinoma and their clinical significance

摘要: 目的 探讨细胞周期蛋白D1(Cyclin D1)、第10号染色体同源丢失性磷酸酶张力蛋白(PTEN)在甲状腺乳头状癌(PTC)组织中的表达情况及临床意义。方法 收集2013年3月至2014年12月手术切除的PTC组织和配对癌旁组织各70例。采用免疫组化En Vision两步法检测上述组织中Cyclin D1、PTEN的表达情况;分析Cyclin D1、PTEN蛋白表达与PTC临床病理特征的关系以及两种蛋白表达的相关性。
结果在70例PTC组织中,Cyclin D1蛋白的阳性表达率为77.1%(54/70),高于癌旁组织的12.9%(9/70),差异有统计学意义(P<0.001);PTEN蛋白的阳性表达率为21.4%(15/70),低于癌旁组织的81.4%(57/70),差异有统计学意义(P<0.001)。Cyclin D1表达与淋巴结转移、临床分期有关(P<0.05),而与年龄、性别、肿瘤大小无关(P>0.05)。PTEN表达与性别、淋巴结转移、临床分期有关(P<0.05),而与年龄、肿瘤大小无关(P>0.05)。Cyclin D1与PTEN蛋白表达无明显相关性(r=0.105,P=0.386)。结论 PTC组织中Cyclin D1高表达,而PTEN低表达,且两者均与淋巴结转移和临床分期有关,提示两者在PTC的发生、发展中发挥一定的作用。

Abstract: Objective To investigate the expression of Cyclin D1 and phosphatase and tensin homolog deleted on chromosome ten (PTEN) in patients with papillary thyroid carcinoma (PTC), and their clinical significance. Methods From March 2013 to December 2014, 70 cases of PTC and paired adjacent normal tissues were enrolled. The expression of PTEN and Cyclin D1 in above tissues was detected by immunohistochemical EnVision twostep method. The correlation of Cyclin D1 and PTEN, as well as the relationship of them with clinicopathological features were analyzed. Results The positive rate of Cyclin D1 was 77.1% (54/70) in PTC tissues, higher than 12.9% (9/70) of adjacent normal tissues with statistical significance (P<0.001). The positive rate of PTEN was 21.4%(15/70)in PTC tissues, lower than 81.4% (57/70) of adjacent normal tissues with statistical sgnificance(P<0.001). The expression of Cyclin D1 was related with lymph node metastasis and clinical staging(P<0.05), but not with age, gender and the size of tumor(P>0.05). The expression of PTEN was associated with gender, lymph node metastasis and clinical staging(P<0.05), but not with age and the size of tumor(P>0.05). Cyclin D1 was not correlated with PTEN (r=0.105, P=0.386). Conclusion Cyclin D1 is overexpressed in PTC tissues, while PTEN is lower expressed. The two proteins are both related with lymph node metastasis and clinical staging. Cyclin D1 and PTEN may play a role in the occurrence and development of PTC.
