The impact of RNA interfering CCDC34 on the biological functions of bladder cancer cells

Objective To evaluate the impact of RNA interfering coiled-coil domain-containing 34(CCDC34)on the proliferation, apoptosis, colony formation and invasion of human bladder cancer cell lines. Methods Bladder cancer 5637 cells were infected with lentivirus-mediated CCDC34 negative control siRNA(negative control group)and CCDC34 siRNA(CCDC34siRNA group)respectively. Fluorescence microscopy was used to observe the infection efficiency. qPCR and Western blotting were used to confirm the interfering efficiency of CCDC34. The cell proliferation,apoptosis,colony formation and invasion ability were analyzed by BrdU incorporation assay,flow cytometry,lowdensity cell colony formation and transwell experiments. Results The lentivirus infection efficiency was above 85% for both CCDC34-siRNA group and negative control group. The mRNA expression of CCDC34siRNA group was greatly reduced compared with negative control group(1.00±0.06 vs. 0.16±0.01,P<0.01),as well as the expression of CCDC-34 protein. Compared with negative control group,cell proliferation was significantly inhibited in the CCDC34-siRNA group (P<0.05);the percentage of 5637 cells in apoptosis phase was also significantly increased in the CCDC34-siRNA group[(28.09±0.39)% vs.(9.50±0.01)%,P<0.01)]. The number of colonies in CCDC34-siRNA group was statistically less than that in negative control group(22±4 vs. 310±5,P<0.01). The cell invasion rate of CCDC34-siRNA group was significantly lower than that of negative control group(0.53±0.03 vs.2.58±0.10,P<0.01). Conclusion Down-regulation of CCDC34 in bladder cancer cell line 5637 via lentivirusmediated siRNA can inhibit the cell proliferation,colony formation and invasion. CCDC34 may serve as a biomarker or even a therapeutic target for bladder cancer.

Reversal effect and mechanism of arsenic trioxide on multidrug resistance of human osteosarcoma cells MG63/dox

Objective To investigate the effect of arsenic trioxide(As2O3)on human osteosarcoma cells MG63 and drug resistant cells MG63/dox,and to explore its possible mechanism. Methods MG63 and MG63/dox cells were respectively treated with adriamycin(ADM, 1-1000 ng/ml)or As2O3(0.25-16 μmol/L) for 48 h; 2 μmol/L As2O3 in combination with ADM(1-1000 ng/ml)was employed to treat MG63/dox cells for 24, 48, 72 h. Meanwhile, negative control group was set. The effect of As2O3 and ADM on proliferation of MG63/dox cell were detected by CCK8 assay. The cell cycle and apoptotic rate of MG63/dox cells after treatment with As2O3(2 μmol/L)and ADM(200 ng/ml)alone and their combination for 48 h were detected by flow cytometry, and the expression levels of P-gp,Bcl-2 and Bax proteins treated by As2O3(2 μmol/L)or ADM(200 ng/ml) alone or in combination for 48 h were analyzed by Western blotting,respectively. Results As2O3 or ADM alone treated MG63/dox cells for 48 h, the proliferation of cells had no obvious changes. The concentration of 4, 8, 16 μmol/L As2O3 and 100, 200, 400, 500, 1000 ng/ml ADM respectively treated MG63 cells for 48 h, the cell proliferation was inhibited(P<0.05). As2O3 in combination with ADM markedly inhibited cell proliferation in MG63/dox cells compared with ADM group and control group(P<0.05)in timedependent manner. Compared with the control group,As2O3 in combination with ADM increased the apoptosis of MG63/dox cells and G2/M phase cell cycle arrest, but reduced G0/G1 phase cell arrest(P<0.05). Furthermore,the expression of Bax protein was found to be upregulated in MG63/dox cells,and the expressions of P-gp and Bcl-2 proteins were lower than those in control group(P<0.05). Conclusion As2O3 combined with ADM can inhibit the proliferation of MG63/dox cells. This effect may associate with induction of apoptosis,up-regulation of Bax expression and down-regulation of P-gp and Bcl-2 expressions.

ObjectiveTo investigate the effect of morphine(Mor)combined with cisplatin(DDP)on the proliferation,apoptosis, as well as invasion and metastasis of human ovarian cancer Skov3 cells. Methods MTT method was used to observe the cell survival rates at 24,48,72,96 h after the treatment of mor at different concentrations(10,20,50,100,200 μmol/L)and DDP(1,2,5,10 and 20 mg/L) as to screen the concentration of Mor and DDP for subsequent functional study. Skov3 cells at logarithmic growth phase were divided into 4 groups:Mor(200 μmol/L)group,DDP(20 mg/L)group,Mor(200 μmol/L)+DDP(20 mg/L) group and control group(Skov3 cells without treatment). Flow cytometry and real-time quantitative PCR were used to detect the apoptosis rate and mRNA levels of apoptosis related genes(Bcl-2,caspase-3 and Bax)at 48 h after the treatment. Transwell test and Western blotting were used to detect the number of cells and the invasion associated proteins(TIMP-2,TIMP-1 MMP-9 and MMP-2)at 48 h after treatment. Results The proliferation inhibition rates increased gradually with the increase of Mor(10-200 μmol/L)and DDP(1-20 mg/L)concentration. The inhibitory effect was in a time and concentration dependent manner. The proliferation inhibition rates of Mor+DDP group was higher than those of Mor group,DDP group and control group at 24, 48,72,96 h after treatment(P<0.05). Compared with control group,there were increased apoptosis rate,caspase-3 activation rate and mRNA levels of Bax and caspase-3, as well as protein levels of TIMP-1 and TIMP-2,but decreased Bcl-2 mRNA,number of cells penetrating membrane and protein levels of MMP-9 and MMP-2 in cells treated with Mor or DDP alone or in combination(P<0.05). The levels of the above indexes in Mor+DDP group were better than those in Mor or DDP groups(P<0.05). Conclusion Both Mor and DDP can inhibit the proliferation of Skov3 cells and induce apoptosis,and the inhibition of Mor combined with DDP is enhanced.

Influence of ALDH1A1 overexpression on biological behavior of gastric cancer cells

Objective To evaluate the effect of overexpression of aldehyde dehydrogenases 1A1(ALDH1A1)on the proliferation,clone formation and invasion of human gastric cancer cell.Methods The eukaryotic expression vector,pEGFP-N1-ALDH1A1,was transfected into gastric cancer cell line MKN-28(experimental group). The empty vector group(MKN-28 cells transfected with empty vector pEGFPN1)and blank control group(MKN-28 cells without any treatment)were also prepared. The methyl thiazolyl tetrazolium(MTT),clone formation assay and Transwell chamber invasion assay were used to evaluate the cell proliferation,clone formation and invasion ability at 96 h after transfection,respectively. Results The proliferation inhibition rate of experimental group was(19.26±2.01)%,lower than that in empty vector group and blank control group. While the clone formation rate and number of cells penetrating membrane were(25.44±1.74)% and 219.78±12.93,higher than those in the empty vector group and blank control group(P<0.05). No significant difference was observed on the above items between blank control group and empty vector group(P>0.05). Conclusion Overexpression of ALDH1A1 can increase the proliferation,colony formation and invasion ability of human gastric cancer cell MKN-28.ALDH1A1 may be involved in the occurence and development of gastric cancer.

Effect of sodium cantharidinate on apoptosis and PUMA expression of colon cancer cell line HCT116

Objective To explore the effect of sodium cantharidinate on the apoptosis and expression of p53upregulated modulator of apoptosis(PUMA)of colon cancer cell line. Methods The colon cancer HCT116 cells were treated with different concentrations of sodium cantharidinate(0.1,0.25,0.5,1,2 μg/ml) for 24 h, and control group without sodium cantharidinate was set. The transepithelial electrical resistance(TER)was analyzed to evaluate the influence of sodium cantharidinate on the apoptosis of HCT116 cell. Real-time fluorescence quantitative PCR and Western blotting were used to detect the changes of mRNA and protein level of PUMA in HCT116 cells. The protein level of PUMA was measured by Western blotting in HCT116 cells treated with inhibitor of nuclear factor(NF)-κB(BAY 11-7082)at concentrations of 10,20,40,80 nmol/L or plasmid for inhibitor of NF-κB kinase β(IKKβ)transfection at concentrations of 0.05,0.1,0.2,0.4,0.8 μg/ml. Results There were decreased TER but increased mRNA and protein level of PUMA in different concentrations of sodium cantharidinate versus control group(P<0.05). Compared with the untreated HCT116 cells,transfection with IKKβ plasmid increased the protein expression of PUMA,and while NF-κB inhibitor reduced the effect of sodium cantharidinate up-regulating the PUMA level with significance difference (P<0.05). Conclusion Sodium cantharidinate may promote the PUMA expression via NF-κB pathway,and therefore induce the apoptosis of HCT116 cells.

Relationship between Ras, BRAF, PIK3CA mutation status and clinicopathological characteristics in colorectal cancer

Objective To understand the mutation status of Ras(K-Ras/N-Ras),BRAF,PIK3CA and its associations with clinicopathological characteristics in colorectal cancer(CRC). Methods Tumor specimens were collected from 200 CRC patients treated in Peking University Cancer Hospital from December 2013 to October 2014. The mutation status of Ras,including K-Ras(exon 2,3 and 4),N-Ras(exon 2,3 and 4),BRAF(exon 15)and PIK3CA(exon 9,20)were detected by direct sequencing. The relationships between the gene mutations and clinicopathological parameters were analyzed. Results Ras mutations were found in 92(46.0%)of 200 patients,including 87 cases(43.5%)of K-Ras,of which occurred mainly in exon 2,and 5 cases of N-Ras. One patient carried KRas mutation and N-Ras mutation simultaneously. BRAF mutations were found in 15 patients(7.5%)and all types of BRAF mutation were V600E. K-Ras and BRAF mutations were mutually exclusive. PIK3CA mutations were found in 9 patients(4.5%),which appeared simultaneously in Ras/BRAF mutation tumors. Also,the mutation rate of Ras(K-Ras/N-Ras)in patients ≥65 years was higher than that in patients <65 years(P<0.05). Other clinicopathological features,such as gender,location of the tumor,histological classification,tumor differentiation,TNM staging,local lymph node metastasis,distant metastasis, and post-operative recurrence and metastasis had no relationship with Ras(K-Ras/N-Ras)gene mutations(P>0.05). BRAF/PIK3CA mutation rates were higher in right side colon tumors(P<0.05). No significant associations were observed between BRAF/PIK3CA mutations and gender,age,histological classification,tumor differentiation,TNM staging,local lymph node metastasis, distant metastasis, and post-operative recurrence and metastasis(P>0.05). Conclusion N-Ras/PIK3CA genes were rarely mutated in Chinese CRC patients. K-Ras/N-Ras mutation were associated with age,while BRAF/PIK3CA mutation were only associated with location of the tumor. Combined detection with K-Ras,N-Ras,BRAF and PIK3CA gene in colorectal cancer patients will provide more reliable basis for clinical targeted therapy.

Investigation on risk factors for breast cancer in Xinjiang

Objective To analyze the risk factors of female breast cancer between Uyghur and Han nationalities in Xinjiang, and to provide guidance or intervention based on risk factors of different nationalities. Methods By adopting 1∶1 casecontrol matched study,it conducted unified questionnaire investigation among selected 400 woman patients with breast cancer(200 Uyghur and 200 Han patients respectively),and 400 health control women(200 Uyghur and 200 Han women respectively),and the risk factors for breast cancer were evaluated by the way of univariateχ2 statsistic analysis and binary Logistic regression analysis. Results Multivariate analysis showed that taking estrogencontaining health products,frequent chicken and roast meat take(≥250 g/week)and benign breast disease history were risk factors for Han patients with breast cancer(P<0.05,OR>1); while older age at menarche(>12 years old),breast feed,consumption of alcohol(≥8 gram per week)and physical exercise were protective factors(P<.05,OR<1). Pregnancy times(>twice),educational background(>high school),benign breast disease history and breast cancer family history were risk factors for Uyghur patients(P<0.05,OR>1); while older age at menarche(>12 years old), breastfeed, and physical exercise were protective factors(P<0.05, OR<1). Conclusion There are differences in risk factors for breast cancer between Uyghur and Han nationalities in Xinjiang. Study on those risk factors can provide references for reasonable, individual prevention strategy.

Differentially expressed genes in residual cervical cancer after radiotherapy screened by cDNA microarray

Objective To identify the differentially expressed genes in residual cervical cancer after radiotherapy by cDNA microarray.
Methods Cervical cancer tissues from 3 patients before radiation and after 50 Gy dose of radiation were tested by cDNA microarray to select differentially expressed genes. The microarray results were validated by quantitative real-time PCR. Results Hierarchic cluster analysis identified that 238 differentiated genes exhibited ≥3.0fold change,among which 111 genes were found to be in persistent up-regulation and 127 in persistent down-regulation after 50 Gy dose of radiation. These genes were involved in cell apoptosis and antophagy, cell energy metabolism, angiogenesis,cell apoptosis,cell cycle regulation,cell signaling,DNA damage repair, cell adhesion,and etc. The expression of CXCL12,CD74,FGF7,COL14A1,CD44 and ATM genes was up-regulated after radiation(P<0.05). Meanwhile,PRC1 and RAD54L were downregulated after radiation(P<0.05). Conclusion The differentially expressed genes identified by cDNA microarray are hoped to provide a foundation for the development of new diagnostic strategies and treatment for radiation resistance in cervical cancer.

Relationship between the expressions of ERCC1 in peripheral blood and tumor tissues of malignancies

ObjectiveTo investigate the correlation between the expression of excision repair cross-complementing gene 1(ERCC1)in paraffin tumor tissue and corresponding blood cells of lung,colon,gastric and breast cancer patients. Methods Real-time quantitative PCR was used to detect ERCC1 mRNA expression of 230 lung cancer,160 colon cancer,125 gastric cancer and 100 breast cancer cases from paraffin tumor tissues and corresponding peripheral venous blood samples. Results There was no significant difference in ERCC1 mRNA expression of tumor tissues and peripheral blood among the four types of cancers(P>0.05). The expression of ERCC1 in tumor paraffin tissues of lung,colon,gastric and breast cancer were much higher than that in corresponding peripheral blood(P<0.05). There was a positive correlation between the expression of ERCC1 mRNA in peripheral blood and cancer tissues(lung cancer:r=0.430,P<0.001;colon cancer:r=0.553,P<0.001;gastric cancer:r=0.583,P<0.001;breast cancer:r=0.501,P<0.001). There was no relation between the ERCC1 expression and the clinical characteristics including age and gender in tumor tissues and peripheral blood of lung, colon and breast cancer. The expression of ERCC1 mRNA in tumor tissue and peripheral blood had no correlation with age in gastric cancer, but it had higher expression in male than in female in tumor tissues. Conclusion The expression levels of ERCC1 mRNA in peripheral blood can indirectly reflect their expression levels in lung,colon,gastric and breast cancer tissues.

Expressions of SATB1,KISS1 in tissue chips of human gastric adenocarcinoma and their clinical significance

Objective To examine the expressions of special AT-rich sequence binding protein-1(SATB1),kisspeptin-1(KISS1)in human gastric adenocarcinoma and investigate their relevance and clinical significance. Methods The expressions of SATB1 and KISS1 were detected in 76 gastric adenocarcinoma and 19 adjacent normal gastric tissues by tissue chip technology and immunohistochemical staining. The correlation of the expressions with clinicalpathological features and prognosis were analyzed.Results The positive rate of SATB1 and KISS1 in gastric adenocarcinoma tissues were 80.3%(61/76) and 14.5%(11/76)respectively. Compared with 10.5%(2/19) and 89.5%(17/19)of normal gastric tissue,the differences were statistical significantly(P<0.05). The SATB1,KISS1 expressions were related to invasive depth, metastasis and TNM staging(P<0.05),but not related to age,sex,tumor diameter and histological differentiation(P>0.05).SATB1 was negatively correlated with KISS1(r=-0.736,P<0.01). The median overall survival(OS)of SATB1 positive and negative patients was 16.8 months(95%CI:10.124-21.876 months)and 42.2 months(95%CI:31.804-52.196 months),and the difference was statistically significant(P<0.05). The median OS of KISS1 positive and negative patients was 45.1 months (95% CI:36.736-53.264 months)and 13.6 months(95% CI:9.971-16.029 months),and the difference was statistically significant(P<0.05). Conclusion SATB1 and KISS1 play important roles in carcinogenesis and progression of gastric adenocarcinoma, which can be regarded as useful biomarkers to evaluate the biological characteristics of gastric adenocarcinoma.

Technique of cervical esophagogastric side-to-side stapled anastomosis in minimally invasive esophagectomy

Objective To evaluate the efficacy of cervical esophagogastric side-to-side stapled anastomosis in minimally invasive resection

of esophageal carcinoma. Methods We retrospectively analyzed the clinical data of 110 patients with esophageal carcinoma underwent completely minimally invasive esophagectomy and left cervical esophagogastromy from March 2014 to March 2015. Sixty-five patients underwent cervical esophagogastromy using circular stapler as control group,and the rest 45 patients were treated by cervical esophagogastric side-to-side stapled anastomosis as experimental group. The anastomosis time,postoperative anastomotic leakage,anastomotic stricture and gastroesophageal reflux were compared between the two groups. Results All the patients underwent minimally invasive resection,and no death happened during perioperative period. The time of anastomosis in experiment group was(16.1±2.5)min,shorter than(27.5±1.9) min of control group(P=0.000). The incidence of anastomotic leakage and gastroesophageal reflux 6 months after operation had no significant difference(P>0.05). The incidence of anastomotic stricture 6 months after operation in experimental group was 4.4%,lower than 24.6% of control group(P=0.005). Conclusion The technique of cervical esophagogastric side-to-side stapled anastomosis can shorten the anastomosis time and reduce the incidence of anastomotic stricture 6 months after operation,and have no tendency to increase anastomotic leakage and gastroesophageal reflux 6 months after operation. It is a satisfactory technique for the treatment of esophageal carcinoma.

Sonographic and CT characteristics of hepatic mass relating to liver involvement in hematologic malignancy

Objective To investigate the sonographic and CT features of hepatic masses caused by liver involvement in hematological

malignancies in order to improve diagnostic acurracy. Methods From April 2012 to May 2015,16 patients with hepatic masses caused by liver involvement in hematologic malignancies were respectively analyzed. Hepatic masses were present in patients with non-Hodgkin's lymphoma(9/16),Hodgkin's disease(2/16),multiple myloma(1/16),acute myeloblastic leukemia(2/16),acute lymphocytic leukemia(1/16)and chronic lymphocytic leukemia(1/16). Results The most frequent sonographic appearance was multiple small focal lesions,and less frequent appearance was single lesions. Most lesions showed regular round,round like or oval shapes,while few lesions showed irregular shapes. The hypoechoic lesions were found in 13 cases,while mixechoic and hyperechoic lesions were found in 1 and 2 cases,respectively. Poor or no color flow signal was found in the lesions.Unenhanced CT showed that multiple focal hypoattenuating lesions were in most cases.Enhanced CT showed no evident enhancement or slight enhancement. More lesions were found in right hepatic lobes than in left lobes. The sonographic and CT features of hepatic masses caused by liver involvement in hematological malignancies,including non-Hodgkin's lymphoma,Hodgkin's disease,multiple myloma,acute and chronic leukemia,are similar. Conclusion Ultrasound and CT show certain featrures in hepatic masses caused by liver involvement in hematological malignancies. The combination of ultrasound and CT is preferred as non-invasive imaging diagnostic method.

Investigation and analysis of the current situation of cancer pain treatment in primary hospitals of Guangzhou

Objective To explore the diagnosis,treatment of cancer pain in primary hospitals of Guangzhou, analyze possible disadvantages,

and provide basis to improve the management of cancer pain. Methods By holding training course of cancer pain standardized treatment of primary hospitals and surveying to all attendees,120 questionnaires were collected and analyzed. Results The mean score in basic knowledge of cancer pain management was 12.2±3.5. There were no significant differences of mean score among doctors,nurses and other medical staff(P>0.05).There was no statistical differences between the mean score of medical staff who participated in cancer pain training course and who did not(P>0.05). The common bad habits of using analgesic drugs were invasive administration and opioids agent inproper use. Barriers to optimal treatment of cancer pain in primary hospitals included insufficient knowledge and education of physicians,concern about side effect of opioids, and shortage of opioids agent. Conclusion Strengthening the training of medical physicians about analgesia,supplying sufficient opioids agent and educating patients is a suitabe way to promote standardized cancer pain management in primary hospitals.

Objective To investigate the value of fiberoptic ductoscopy-guided intraductal biopsy in combination with the detection of

biomarker CA153 in diagnosis of early ductal pathological changes. Methods From May 2012 to October 2014,fiberoptic ductoscopy was performed on 123 nipple discharge patients, and patients diagnosed as breast intraductal neoplasm were given intraductal biopsy. The biopsy results were compared with pathology of duct excision. Nipple lavage fluid were collected from patients with intraductal neoplasm for the test of CA153,and 30 patients of intraductal dilatation were enrolled as control. Results Fifty-six patients who were found brest intraductal neoplasm among 123 patients underwent 64 times of fiberoptic ductoscopy-guided intraductal biospy and ductal lesions were removed. Among 56 patients underwent fiberoptic ductoscopy-guided intraductal biopsy,there were intraductal papilloma in 28 cases,mutiple intraductal papilloma in 7 cases,ductal hyperplasia in 6 cases,severe atypical ductal hyperplasias in 2 cases,and breast cancer was found in 7 cases. Biopsy failed in 6 patients, and the success rate of biopsy was 89.3%. All the patients underwent breast duct dissection. There were solitary intraductal papilloma in 29 cases,multiple intraductal papilloma in 11cases,ductal hyperplasia in 8 cases,atypical ductal hyperplasia in 1 case,intraductal carcinoma in 4 cases,invasive ductal carcinoma in 2 cases and intraductal papilloma carcinoma in 1 case. CA153 level of nipple lavage fluid in intraductal dilatation patients was(26.90±2.76)U/ml,lower than(31.91±22.95)U/ml of G3 grade(P>0.05),(52.67±33.63)U/ml of G4(P<0.05)and(85.68±21.13)U/ml of G5 grade(P<0.05). Conclusion Fiberoptic ductoscopy-guided intraductal biopsy is microinvasive,safe,convenient with high success rate,and in combination with CA153 level in nipple lavage fluid shows certain values in diagnosing early ductal malignant changes.

Misdiagnosis of 15 lung cancer patients with digestive tract symptoms as first manifestation

Objective To explore clinical characteristics and misdiagnosis of lung cancer with digestive tract symptoms as first manifestation in order to improve the awareness of the disease. Methods Fifteen primary lung cancer patients with digestive tract symptoms as first manifestation from January 2005 to January 2013 were enrolled,then their clinical data were analyzed retrospectively. Results The first clinical manifestations of 15 primary lung cancer patients included nausea and vomiting(7 cases),anorexia and weight lose(3 cases),eating choking(2 cases),abdominal pain(2 cases),and diarrhea(1 case).There were 9 patients with refractoriness hyponatremia. There were 7 misdiagnosis cases of acute gastritis,3 of chronic gastritis,2 of esophagitis,1 of peptic ulcer,1 of cholelithiasis and 1 of acute enteritis.Eight patients were diagnosed as small cell lung cancer,including 7 cases with syndrome of inappropriate antidiuretic hormone secretion(SIADH)and 1 case with carcinoid syndrome. Two patients were diagnosed as lung squamous cell carcinoma with mediastinal lymph node metastasis. There were 5 patients diagnosed as lung adenocarcinoma,including 2 cases with supraclavicular lymphnode metastasis,2 cases with SIADH and 1 case with brain metastasis. Conclusion Lung cancer with digestive track symptoms as first manifestation is easily misdiagnosed. Clinicians should improve the awareness of primary lung cancer with digestive tract symptoms as first manifestation, and pay more attention to examination and diagnosis.

Treatment and prognosis of 7 cases with sinonasal neuroendocrine carcinoma

Objective To explore the treatment and prognosis of sinonasal neuroendocrine carcinoma(SNEC). Methods A retrospective analysis of 7 patients with SNEC from May 2012 to July 2014 was performed. All the patients were male and age from 39 to 61. The main symptoms were nasal obstruction,headache, and facial swelling and pain. Three patients were in IVA stage,three were in IVB stage and one unknown. One patient underwent preoperative chemotherapy and surgery,one with preoperative chemoradiotherapy and surgery,and five patients received radical radiotherapy in combination with chemotherapy. Results Partial remission was observed in all 7 patients after treatment and complete remission was observed in 2 patients during follow-up. The follow-up ranged from 10.56 months. One patient died of distant metastasis,and the remaining 6 patients were still alive in followup.The side effects included oral mucositis,myelosuppression and transaminase elevation. ConclusionSNEC is rare and highly malignant in clinic without typical clinical presentation. Comprehensive treatment based on radiotherapy may improve prognosis of patients with local advance.

Progress on the mechanism of exosomes in gastric cancer

Exosomes are membrane vesicle-like structures of 30-100 nm in diameter,existing in various body fluids,which are released from a variety of different cell types. Exosomes have emerged as a noval mode of intracellular communication,participating in the progression of oncogenic processes. Gastric cancer derived exosomes can regulate PI3K/Akt,and MAPK/ERK signaling pathways by shuttling key molecules including miRNA,cytokines and protiens,which may influence tumor proliferation,invasion,metastasis and immune escape in gastric cancer. In this review,we discuss the mechanism of exosomes in gastric cancer, which may provide new ideas on diagnosis and treatment of gastric cancer.

Recently,high expression of long non-coding RNA urothelial carcinoma antigen 1(UCA1)has been found in various human carcinomas(bladder urothelial carcinoma,gastric cancer,hepatocellular carcinoma,ovarian cancer,etc)acting as proto-oncogene. UCA1 is playing an important role in the development and progression of tumors. As a novel biological marker,it is indispensible during the process of diagnosis,therapy and prognosis. In this view,we summarize the latest progress,both domestic and overseas,on the advance of UCA1 in tumor research.
